Parathyroid increase with age associated with worsening Vitamin D genes – April 2020

Older Age Is Associated With Decreased Levels of VDR, CYP27B1, and CYP24A1 and Increased Levels of PTH in Human Parathyroid Glands

Vitamin D blood test misses a lot
in Visio for 2023

  • Vitamin D from coming from tissues (vs blood) was speculated to be 50% in 2014, and by 2017 was speculated to be 90%
  • Note: Good blood test results (> 40 ng) does not mean that a good amount of Vitamin D actually gets to cells
  • A Vitamin D test in cells rather than blood was feasible (2017 personal communication)   Commercially available 2019
    • However, test results would vary in each tissue due to multiple genes
  • Good clues that Vitamin D is being restricted from getting to the cells
    1) A vitamin D-related health problem runs in the family

especially if it is one of 51+ diseases related to Vitamin D Receptor

+2) Slightly increasing Vitamin D shows benefits (even if conventional Vitamin D test shows an increase) +3) DNA and VDR tests - 120 to 200 dollars $100 to $250 +4) PTH bottoms out ( shows that parathyroid cells are getting Vitamin d)

Genes are good, have enough Magnesium, etc.

+4) Back Pain

probably want at least 2 clues before taking adding vitamin D, Omega-3, Magnesium, Resveratrol, etc

      • The founder of VitaminDWiki took action with clues #3&4

Items in both categories Thyroid/para and VDR are listed here:

Vitamin D Receptor is associated in over 58 autoimmune studies
The risk of 44 diseases at least double with poor Vitamin D Receptor as of Oct 2019

Vitamin D Receptor activation can be increased by any of: Resveratrol,  Omega-3,  MagnesiumZinc,   Quercetin,   non-daily Vit D,  Curcumin, intense exercise,   Ginger,   Essential oils, etc  Note: The founder of VitaminDWiki uses 10 of the 12 known VDR activators

Parathyroid glands contain the vitamin D receptor (VDR) and 25-hydroxyvitamin D-1α-hydroxylase (CYP27B1) and 24-hydroxylase (CYP24A1), which catalyze the production and degradation of 1,25-dihydroxyvitamin D [1,25(OH)2D], respectively. Previous studies have shown that the serum level of intact parathyroid hormone (iPTH) increases with age. We hypothesized that the expression of CYP27B1 or VDR in parathyroid glands decreases with age, which might account for the increased serum levels of iPTH due to decreased suppression of parathyroid hormone (PTH) secretion by 1,25(OH)2D in older people. To test this hypothesis, we examined relative expression levels of VDR, CYP27B1, CYP24A1, and PTH in specimens from parathyroid glands unintentionally removed during thyroidectomy for 70 patients varying in age from 10 to 70 years. The results showed that there was an inverse correlation between age and VDR, CYP27B1, and CYP24A1 expression (p < 0.05). A significant positive correlation between PTH expression levels and age was also observed (p < 0.05).

These data indicate that older age is associated with decreased levels of VDR, CYP27B1, and CYP24A1 and increased levels of PTH in human parathyroid glands.
